Structural proteomics of protein-protein and protein-drug interactions

 

Abstract

Combining protein chemistry methods with modern mass spectrometry has led to the emergence of the distinct field of structural proteomics. Multiple protein chemistry approaches, such as, surface modification, limited proteolysis, hydrogen–deuterium exchange, cross-linking and photo-affinity labeling, provide unique structural information on the protein systems studied. Experimental data from these various structural proteomics techniques offer valuable structural insights, especially in cases, where traditional structural biology methods, such as, X-ray crystallography, EM, NMR and, recently, machine learning-assisted homology modeling, fall short. Here, we will discuss method development and applications of various structural proteomics approaches for studying of protein structure, protein-protein and protein-drug interactions.

 

 

Biosketch

Evgeniy V. Petrotchenko received his M.D. from the Second Moscow Medical Institute, Moscow, Russia and his Ph.D. in bioorganic chemistry from the Institute of Bioorganic Chemistry of the National Academy of Sciences of Belarus, Minsk, Belarus. He conducted post-doctoral studies at the National Institute of Environmental Health Sciences, National Institutes of Health in Research Triangle Park, NC, USA and at the University of North Carolina at Chapel Hill, NC, USA. He joined Christoph Borchers lab as Research Associate at UNC-CH and, later, as Research Assistant Professor at the University of Victoria-Genome BC Proteomics Centre, University of Victoria, BC, Canada and as Associate Director of the Segal Cancer Proteomics Centre, Lady Davis Institute, Montréal, QC, Canada. He is co-founder of Creative Molecules Inc. and of the Symposium on Structural Proteomics.

Main research focus is structural proteomics method development and applications and, recently, metabolomics and clinical MS.
