Label-Free LC-MS/MS Analysis of Reactive Metabolite Protein Targets
Liver plays a predominant role in metabolic transformations and hepatic proteins are often targeted by reactive metabolites potentially leading to hepatotoxicity. Knowing what proteins are targeted is important to understand the mechanisms involved in this type of toxicity. An analytical approach was developed to identify reactive metabolite protein targets and their sites of modification using LC-MS/MS. This research was conducted in three phases to address the main challenges posed in this type of analysis: sample preparation, LC-MS/MS detection, and data analysis. The developed workflow was applied to conduct the first reliable identification and site-mapping of acetaminophen liver protein adducts from a complex proteome system by a global analysis approach.
