Investigating Heterogeneity and Functional Niches of Macromolecular Machines

 

Abstract

After transcription, all RNA species are assembled into ribonucleoprotein (RNP) complexes to be matured via different pathways defined by specific proteins that form discrete subsets associating with each RNA in a dynamic fashion. Yet even for a specific RNA specie these subsets can differ, either due to alternative maturation pathways for specialized niches of gene expression, cellular perturbations, cell types, tissues, or disease. We are developing and applying proteomic approaches to study these different aspects of RNA metabolism. Combining affinity purification with sqMS, crosslinking, isotope labeling as well as DIA, we investigate interactome variations of heterogeneous and spatiotemporal RNP composites.

 

Biosketch

Dr. Marlene Oeffinger is an Associate Research Professor at the IRCM, where she started her lab in December 2009, and Professeure agregée in the département de biochimie et médecine moléculaire at the Université de Montréal. Her group’s work is centered around determining the spatial arrangement, stoichiometry, and substrate specificity that define heterogeneity of ribonucleoprostein complexes using and developing proteomic, RNomic and biochemical approaches. She has received funding from the CIHR, NSERC, ALS Canada and FRQ-S, and has published 31 peer-reviewed articles and one book to date.
